What Is Epitalon?

Published

Epitalon, also spelled epithalon or epithalone, is a synthetic tetrapeptide of 4 amino acids: Ala-Glu-Asp-Gly (AEDG). It was synthesized from the amino-acid composition of epithalamin, a bovine pineal gland extract. A 2025 review counted about 25 years of in vitro, in vivo and in silico work on it. The best-known findings are telomerase and telomere measurements in cultured human cells and lifespan and tumor counts in rodents, much of it from a small number of research groups.

Where epitalon comes from

Epitalon is the tetrapeptide Ala-Glu-Asp-Gly, abbreviated AEDG and also written as epithalon or epithalone. A 2025 review describes it as synthesized on the basis of the amino-acid composition of epithalamin, a polypeptide extract of bovine pineal gland, before it was itself found in a pineal gland polypeptide complex solution.Review · PMID 40141333

The same review notes an imbalance in the literature. Over the last 25 years the compound has been studied with in vitro, in vivo and in silico methods, and many biological effects have been reported, yet physico-chemical and structural studies of the peptide itself remain quite limited. The review also states that it remains uncertain which mechanisms account for the reported effects.Review · PMID 40141333

Effects the review lists from earlier studies include an influence on melatonin synthesis, changes in interleukin-2 mRNA levels, modulation of the mitogenic activity of murine thymocytes, and changes in the activity of enzymes including AChE, BuChE and telomerase. These are reported as findings from the literature it surveys, not as new measurements.Review · PMID 40141333

Telomerase and telomeres in cultured human cells

The study most often cited is a short 2003 report. Adding epithalon to a culture of telomerase-negative human fetal fibroblasts induced expression of the catalytic subunit of telomerase, telomerase enzyme activity and telomere elongation. The authors attributed this to reactivation of the telomerase gene in somatic cells. In vitro · PMID 12937682

A 2025 study from a different group repeated the question in more cell types: the breast cancer lines 21NT and BT474, and normal epithelial and fibroblast cells. Using qPCR and immunofluorescence, it reported telomere length extension in normal cells that rose with concentration and ran through hTERT and telomerase upregulation. In vitro · PMID 40908429

In the two cancer cell lines, the same study also recorded significant telomere length extension, but through ALT, the alternative lengthening of telomeres pathway, rather than through telomerase. ALT activity was not observed in the normal cells, so the authors described it as specific to the cancer cells. The study did not test whether telomere changes in culture affected any function of the cells or tissues they came from.In vitro · PMID 40908429

Other cell and oocyte models

In a 2025 study of a human retinal pigment epithelial cell line, ARPE-19, high glucose delayed closure in a scratch-wound assay and raised intracellular reactive oxygen species. It also induced epithelial-mesenchymal transition and fibrosis-related genes. In this model, scratch-wound closure in the high-glucose cells returned toward that of control cells with epitalon, which also inhibited the glucose-induced epithelial-mesenchymal transition and fibrosis changes. The authors called for more mechanistic and safety work.In vitro · PMID 40493162

A 2022 study examined mouse oocytes kept in culture after ovulation, a model in which egg quality declines with time. Over 6, 12 and 24 hours of culture, epitalon in the medium was associated with lower intracellular reactive oxygen species. At 12 and 24 hours it was also associated with fewer spindle defects and less abnormal cortical granule distribution, along with higher mitochondrial membrane potential and mitochondrial DNA copy number. Oocyte apoptosis at 24 hours was lower. In vitro · PMID 35413689

A 2022 study of the human THP-1 monocyte line tested epitalon alongside four other short peptides. All five changed proliferative signaling, increasing tyrosine phosphorylation of mitogen-activated kinases, and all inhibited LPS-stimulated expression of TNF and IL-6 in differentiated THP-1 cells. The design did not isolate an effect unique to epitalon. In vitro · PMID 35408963

Lifespan and tumor studies in rodents

In a 2001 study, female CBA mice received synthetic epithalon from 6 months of age until death. The authors reported no effect on body weight, food consumption, physical activity or behavior. They did report a slower age-related decline of estrous function, lower body temperature, slower free-radical processes, a longer lifespan and a lower incidence of spontaneous tumors.Animal study · PMID 11227856

Two later rat studies tested the peptide under different light regimes. In female rats, constant or natural light shortened mean lifespan by 13.5% and 25.5%. Epithalon did not change lifespan under a standard day/night regime, but under natural and constant light, maximum lifespan was 95 and 24 days longer, and spontaneous tumors were inhibited only under natural light.Animal study · PMID 18856211

In male rats, epithalone given from 4 months of age until natural death virtually did not change mean lifespan. It was associated with a significant (p<0.05) normalization of the population aging rate under natural or constant light, and in every light regime it inhibited spontaneous tumors, mainly testicular leydigomas and leukemias.Animal study · PMID 19110597

A 2004 analysis took a different approach. Rather than reporting a single survival figure, it fitted a semi-parametric model of heterogeneous mortality to survival data from female HER-2/neu transgenic mice exposed to several stressors, including light deprivation and constant light, and to epitalon or melatonin. The study was designed so that the effects of these different conditions on survival could be compared within one framework. The model separates changes in population heterogeneity from changes in underlying hazard, and the authors used it to describe effects such as debilitation and hormesis rather than to estimate a single lifespan difference.Animal study · PMID 15501020

How to read the epitalon evidence

The rodent results are not uniform. Mean lifespan in male rats was virtually unchanged, and in female rats lifespan was unchanged under standard light. Differences appeared mainly under altered light regimes, and the tumor findings depended on sex and light condition.Animal study · PMID 18856211Animal study · PMID 19110597

The telomere findings come from cell culture. The 2003 study used one telomerase-negative fibroblast culture, and the 2025 study, while broader, found a different mechanism in cancer lines than in normal cells. Neither measured anything beyond the cultured cells. In vitro · PMID 12937682In vitro · PMID 40908429

The 2025 review concludes that, despite the volume of biological research, it is still uncertain whether the reported mechanisms are the only ones, and structural work on the peptide remains limited. No randomized trial of epitalon in people is among the studies summarised here.Review · PMID 40141333
